MakConkey Agar, laktozu fermente eden ve pH düşüşüyle pembe-kırmızı renge dönen gram-negatif organizmaların izolasyonu için kullanılır. Safra tuzlarının varlığı gram-pozitif organizmaların büyümesini engeller, bu nedenle bu agarda üreme, özellikle BOS kültüründe CNS enfeksiyonunu düşündürür.
