Antibody Antigen Slide Unattached Washed Added Binds Enzyme

The ELISA test involves applying a sample to a slide to allow antigens to attach, washing away unbound antigens, adding a specific antibody that binds to the target antigen, washing again, adding an enzyme-linked antibody that binds to the first, and finally adding a substrate that produces a measurable color change.

- sample applied to slide - antigens attach to slide - unattached antigens washed off - antibody complementary to antigen being tested for added - binds to antigen - unattached antibodies washed off - second antibody attached to enzyme added - binds to first antibody - colourless substrate added - enzyme acts on substrate - coloured product produced - intensity of colour measured to determine antigen concentration (antigens, antibody 1, antibody 2 with enzyme, substrate)

Front How does the ELISA test work?
Back - sample applied to slide
- antigens attach to slide
- unattached antigens washed off

- antibody complementary to antigen being tested for added
- binds to antigen
- unattached antibodies washed off

- second antibody attached to enzyme added
- binds to first antibody

- colourless substrate added
- enzyme acts on substrate
- coloured product produced
- intensity of colour measured to determine antigen concentration
