Bases Start Point End Numbers Left Are Upstream The 5

Bases located upstream (towards the 5' end) of the start point are given negative numbers, while bases downstream (towards the 3' end) are given positive numbers.

Bases before the start point (5' end) are upstream and negative. Bases after the start point (3' end) are downstream and positive.
