crystal violet applied with iodine (fixes dye) onto specimen and washed with alcohol, gram-positive retains stain (so blue/purple), gram-negative have thinner cell walls so lose stain, (so stained with a counterstain, safranin dye, and appear red), gram-positive susceptible to antibiotics as they inhibit formation of cell walls
