    • wire inoculating loop sterilised by holding close to flame until glows red hot
    • cool and dip in bacterial suspension
    • remove petri dish lid and streak loop horizontally across agar surface in zig-zag pattern
    • replace lid and hold with tape (not fully sealed to allow O2 to enter and prevent formation of anaerobic bacteria) and incubate at set temp
