Number Factor Method Effect Growth Experiment Dilution Original

The method to investigate the effect of one factor on microorganism growth involves aseptic techniques and serial dilution to count colonies on agar plates. The colony count, assumed to originate from a single microorganism, is used to determine the initial bacterial numbers, which are then monitored for population changes after the investigated factor is applied, while keeping other factors constant.

  • Ensure aseptic conditions throughout experiment
  • Carry out serial dilution of original culture broth until when culturing a given volume of broth on an agar plate, the number of colonies can be counted 
  • Assuming that each colony on a dilute agar plate comes from a single microorganism, the number of colonies indiciates the number of bacteria that was initially on the dilute plate
  • By multiplying this number by the dilution factor, the cell count per volume of original colony can be found
  • This method is carried out at start of experiment to count initial number of bacteria and at the end, after the desired factor has affected the population
