Back - formed in red bone marrow, enter circulation without nuclei (so only lasts 120 days), contains haemohlobin (red pigment carrying O2) which is globular conjugated protein made of 4 peptide chains (each with a haem group) so each haemoglobin can bind to 4 O2 loosely to form oxyhaemoglobin 

- when entering caps in lungs, O2 levels low so steep conc gradient between inside of RBC and air in alveoli, so O2 moves in and binds either Hb, as soon as a O2 binds to haem group, RBC change shape so it’s easier for next O2 to bind (positive cooperativity), as O2 bound to Hb, steep conc gradient maintained between inside RBC and air until all hemoglobin saturated 

- if at body tissues, opp occurs as conc of O2 in cytoplasm of cells Lower than in RBC, so O2 moves out, changing shape of RBC, so easier to lose rest of O2
